Plaque reduction-based Antiviral Screening Assay Services.
The Viral Bioassay facility (VBaf) offers antiviral assay testing services to screen the efficacy of your test compounds to inhibit viral infections. The assay is based on the Plaque Reduction Neutralization Test (PRNT) which measures the ability of the test compounds to neutralize a virus. The compounds already validated to show antiviral activity against the virus of interest will be used as an internal control.
Features of Antiviral assay
- Qualitative Cytotoxicity testing
- 4-point dilution Plaque reduction analysis in duplicates
The assay format is given below:
Compound screening
Requirements
- The test compound should be soluble in water, alcohol or DMSO at a minimum concentration of 1mM (for pure molecules) or 1mg/ ml (for crude extract).
- The antiviral activity of the test compound shall be tested in Baby Hamster Kidney fibroblast cells (BHK-21 line) or Vero cells, hence the test compound should be nontoxic to these cells.
- The solubilized test compound should remain in solution in the cell culture medium (DMEM+ 2% FBS) at the final concentration of 0.01μM or 10 μM (for pure molecules) and 10μg or100 μg for crude extracts.
Available tests
- Anti Chikungunya virus
- Anti Dengue virus
- Anti Respiratory Syncytial Virus
